+1 voteMelbourne Zoo - having been a world traveler I'm proud that Melbourne's zoo is still among the best in the world.
Scienceworks - It's a science museum
Melbourne Museum - It's relatively new.
As another poster recommended the MCG's Sports Museum is outstanding.
Federation Square - in the heart of the city. Is home to national gallery and some ACME.
Go to an AFL game if you're interested in sports, preferably at the MCG. That's pure Melbourne culture there.

Just came back from Melbourne. With 2 kids definitely Scienceworks, Federation Square (which currently has Game Masters on) and Melbourne Museum.

Probably Brunswick St in Fitzroy for street clothes shopping, Chapel for slightly more upmarket and the city is pretty decent for certain things, try little collins/melbourne central
